ABSTRACT: SUMMARY Meta-analyses of genome-wide association studies (GWAS) have identified >240 loci associated with type 2 diabetes (T2D), however most loci have been identified in analyses of European-ancestry individuals. To examine T2D risk in East Asian individuals, we meta-analyzed GWAS data in 77,418 cases and 356,122 controls. In the main analysis, we identified 301 distinct association signals at 183 loci, and across T2D association models with and without consideration of body mass index and sex, we identified 61 loci newly implicated in T2D predisposition. Common variants associated with T2D in both East Asian and European populations exhibited strongly correlated effect sizes. New associations include signals in/near GDAP1, PTF1A, SIX3, ALDH2, a microRNA cluster, and genes that

Lifestyle context

Concrete actions anchored to the cited research. We do not prescribe, we describe.

Diet

  • Whole grains and limited refined carbohydrates Moderate

Dietary quality affects glucose metabolism; variant confers increased T2D susceptibility warranting carbohydrate quality emphasis

Emphasize whole grains and legumes; limit refined carbohydrates and added sugars

Exercise

  • Regular physical activity for type 2 diabetes prevention Moderate

Physical activity is key modifiable factor for type 2 diabetes risk; variant increases susceptibility warranting prevention emphasis

150+ minutes moderate-intensity aerobic activity per week

Lifestyle

  • Maintain healthy weight for type 2 diabetes prevention Moderate

Body weight strongly influences insulin sensitivity and type 2 diabetes risk; elevated genetic risk emphasizes weight management importance

Target BMI 18.5-24.9 or discuss goal weight with physician

Screening

  • Type 2 diabetes screening Moderate

Variant rs12109081 T allele is associated with ~4% increased type 2 diabetes risk per allele

Baseline screening and repeat every 2-3 years; discuss optimal frequency with physician
