  • High Blood Pressure and Intraocular Pressure: A Mendelian Randomization Study - Unknown journal (n.d.) · Unknown authors · PubMed 35762941

ABSTRACT: Purpose To test for causality with regard to the association between blood pressure (BP) and intraocular pressure (IOP) and glaucoma. Methods Single nucleotide polymorphisms (SNPs) associated with BP were identified in a genome-wide association study (GWAS) meta-analysis of 526,001 participants of European ancestry. These SNPs were used to assess the BP versus IOP relationship in a distinct sample (n = 70,832) whose corneal-compensated IOP (IOPcc) was measured. To evaluate the BP versus primary open-angle glaucoma (POAG) relationship, additional Mendelian randomization (MR) analyses were conducted using published GWAS summary statistics. Results Observational analysis revealed a linear relationship between BP traits and IOPcc, with a +0.28 mm Hg increase in IOPcc per 10-mm Hg inc

  • Participation bias in the UK Biobank distorts genetic associations and downstream analyses - Unknown journal (n.d.) · Unknown authors · PubMed 37106081

ABSTRACT: While volunteer-based studies such as the UK Biobank have become the cornerstone of genetic epidemiology, the participating individuals are rarely representative of their target population. To evaluate the impact of selective participation, here we derived UK Biobank participation probabilities on the basis of 14 variables harmonized across the UK Biobank and a representative sample. We then conducted weighted genome-wide association analyses on 19 traits. Comparing the output from weighted genome-wide association analyses (neffective = 94,643 to 102,215) with that from standard genome-wide association analyses (n = 263,464 to 283,749), we found that increasing representativeness led to changes in SNP effect sizes and identified novel SNP associations for 12 traits. Lifestyle context

Concrete actions anchored to the cited research. We do not prescribe, we describe.

Diet

  • DASH-style diet pattern Moderate

DASH diet reduces blood pressure and LDL cholesterol, the two traits elevated by rs11852760-A

Emphasize vegetables, fruits, whole grains, lean proteins; limit sodium and saturated fat

Exercise

  • regular cardiovascular exercise Moderate

Aerobic activity mitigates genetic predisposition to elevated diastolic blood pressure linked to SEMA7A

150 min moderate intensity per week, or 75 min vigorous intensity

Screening

  • annual blood pressure check Moderate

rs11852760-A increases diastolic blood pressure (effect 0.23 mmHg per allele, n=526,001, p=2e-27)

Annual measurement; baseline if not recently checked

  • lipid panel for LDL cholesterol Moderate

    rs11852760-A increases LDL cholesterol (effect 0.014-0.018 mmol/L, n=97k-269k, p<1e-5)

Baseline lipid panel; repeat every 5 years if normal, annually if elevated
