Identification of type 2 diabetes loci in 433,540 East Asian individuals - Unknown journal (n.d.) · Unknown authors · PubMed 32499647
ABSTRACT: SUMMARY Meta-analyses of genome-wide association studies (GWAS) have identified >240 loci associated with type 2 diabetes (T2D), however most loci have been identified in analyses of European-ancestry individuals. To examine T2D risk in East Asian individuals, we meta-analyzed GWAS data in 77,418 cases and 356,122 controls. In the main analysis, we identified 301 distinct association signals at 183 loci, and across T2D association models with and without consideration of body mass index and sex, we identified 61 loci newly implicated in T2D predisposition. Common variants associated with T2D in both East Asian and European populations exhibited strongly correlated effect sizes. New associations include signals in/near GDAP1, PTF1A, SIX3, ALDH2, a microRNA cluster, and genes that

Lifestyle context
Concrete actions anchored to the cited research. We do not prescribe, we describe.
Diet
-
refined carbohydrates and added sugars Moderate
Refined carbs cause rapid glucose spikes; ST6GAL1 variants impair glucose homeostasis
limit processed foods, choose whole grains, aim for <25g added sugars daily
Discuss with your doctor
-
type 2 diabetes risk discussion and prevention planning Moderate
Clinical risk stratification optimizes preventive intervention intensity
Exercise
-
progressive resistance training Moderate
Muscle training enhances insulin sensitivity and metabolic capacity
2+ sessions per week, major muscle groups
-
regular moderate-intensity aerobic exercise Moderate
Aerobic exercise improves insulin sensitivity and glucose clearance
150+ minutes per week at moderate intensity
Lifestyle
-
maintain healthy body weight or achieve modest weight loss Moderate
Excess adiposity worsens insulin resistance; modest weight loss reduces diabetes risk
if overweight, target 5-10% weight loss
Screening
-
annual fasting glucose and hemoglobin A1c screening Moderate
Early detection enables intervention before diabetes progression
annual HbA1c and fasting glucose
