ABSTRACT: Metabolic syndrome (MetS) is a complex hereditary condition comprising various metabolic traits as risk factors. Although the genetics of individual MetS components have been investigated actively through large-scale genome-wide association studies, the conjoint genetic architecture has not been fully elucidated. Here, we performed the largest multivariate genome-wide association study of MetS in Europe (nobserved = 4,947,860) by leveraging genetic correlation between MetS components. We identified 1,307 genetic loci associated with MetS that were enriched primarily in brain tissues. Using transcriptomic data, we identified 11 genes associated strongly with MetS. Our phenome-wide association and Mendelian randomization analyses highlighted associations of MetS with diverse di

Lifestyle context

Concrete actions anchored to the cited research. We do not prescribe, we describe.

Bloodwork

  • Annual monitoring of metabolic parameters Moderate

Regular assessment of glucose homeostasis and lipid metabolism enables early detection of metabolic dysfunction

Annual fasting glucose, lipid panel, blood pressure, waist circumference; more frequent if abnormalities detected

Diet

  • Mediterranean or DASH-style diet Moderate

These dietary patterns reduce glucose intolerance, dyslipidemia, and adiposity; core metabolic syndrome features

Emphasize whole grains, vegetables, legumes, olive oil, fish; minimize refined carbohydrates and added sugars

Exercise

  • Regular aerobic and resistance training Moderate

Physical activity reduces metabolic syndrome components; genetic predisposition increases benefit from preventive exercise

150 minutes moderate aerobic activity + 2 days/week resistance training

Screening

  • Metabolic syndrome screening (every 5 years) Moderate

rs1107392 INKA2 variant is associated with increased metabolic syndrome risk; screening enables early intervention

Fasting glucose, lipid panel, blood pressure, waist circumference measurements per provider guidelines
