Diversity and scale: Genetic architecture of 2068 traits in the VA Million Veteran Program - Unknown journal (n.d.) · Unknown authors · PubMed 39024449
ABSTRACT: INTRODUCTION: Findings from genome-wide association studies (GWASs) have provided foundational knowledge of the genetic basis of disease, facilitating precision approaches for prevention and treatment. Current GWAS results are limited by underrepresentation of individuals from diverse populations, leading to concerns with generalizability regarding our knowledge of the relationships between genes, traits, and disease. The Department of Veterans Affairs (VA) Million Veteran Program (MVP), one of the largest US-based biobanks, addresses this need; 29% of MVP comprises individuals genetically similar to African (AFR), Admixed American (AMR), and East Asian (EAS) reference populations. With over 635,000 participants and more than 44.3M genotyped variants linked with detailed phenotyp

Identification of type 2 diabetes loci in 433,540 East Asian individuals - Unknown journal (n.d.) · Unknown authors · PubMed 32499647
ABSTRACT: SUMMARY Meta-analyses of genome-wide association studies (GWAS) have identified >240 loci associated with type 2 diabetes (T2D), however most loci have been identified in analyses of European-ancestry individuals. To examine T2D risk in East Asian individuals, we meta-analyzed GWAS data in 77,418 cases and 356,122 controls. In the main analysis, we identified 301 distinct association signals at 183 loci, and across T2D association models with and without consideration of body mass index and sex, we identified 61 loci newly implicated in T2D predisposition. Common variants associated with T2D in both East Asian and European populations exhibited strongly correlated effect sizes. New associations include signals in/near GDAP1, PTF1A, SIX3, ALDH2, a microRNA cluster, and genes that

Lifestyle context
Concrete actions anchored to the cited research. We do not prescribe, we describe.
Diet
-
Dietary emphasis on whole grains and plant-based foods High
Whole grain and fiber intake is inversely associated with Type 2 diabetes risk
Emphasize whole grains, legumes, vegetables, nuts; limit refined grains and added sugars
Exercise
-
Regular physical activity for Type 2 diabetes prevention High
Aerobic exercise improves insulin sensitivity and glucose homeostasis in genetic T2D risk
150 minutes moderate-intensity aerobic activity per week plus 2-3 sessions resistance training
Lifestyle
-
Maintain body weight in normal range (BMI 18.5-24.9) High
Excess adiposity impairs insulin sensitivity and elevates Type 2 diabetes risk
Monitor weight regularly; maintain BMI below 25 through diet and exercise
Screening
-
Type 2 diabetes screening beginning at age 30 High
rs10852123 A allele increases Type 2 diabetes risk 1.06-fold per copy
Annual fasting glucose or HbA1c screening starting at age 30; earlier if other risk factors present
