We performed a genome-wide association meta-analysis (GWAMA) of 290,134 attention-deficit/hyperactivity disorder (ADHD) symptom measures of 70,953 unique individuals from multiple raters, ages and instruments (ADHD ). Next, we meta-analyzed the results with a study of ADHD diagnosis (ADHD ). ADHD returned no genome-wide significant variants. We show that the combined ADHD GWAMA identified 39 independent loci, of which 17 were new. Using a recently developed gene-mapping method, Fine-mapped Locus Assessment Model of Effector genes, we identified 22 potential ADHD effector genes implicating several new biological processes and pathways. Moderate negative genetic correlations (r < -0.40) were observed with multiple cognitive traits. In three cohorts, polygenic scores (PGSs) based on ADHD ou

  • Genetic architecture reconciles linkage and association studies of complex traits. - Nature genetics (2024) · Sidorenko J, Couvy-Duchesne B, Kemper KE, Moen GH, Bhatta L, Åsvold BO, Mägi R, Ani A, Wang R, Nolte IM, Gordon S, Hayward C, Campbell A, Benjamin DJ, Cesarini D, Evans DM, Goddard ME, Haley CS, Porteous D, Medland SE, Martin NG, Snieder H, Metspalu A, Hveem K, Brumpton B, Visscher PM, Yengo L · PubMed 39375568

Linkage studies have successfully mapped loci underlying monogenic disorders, but mostly failed when applied to common diseases. Conversely, genome-wide association studies (GWASs) have identified replicable associations between thousands of SNPs and complex traits, yet capture less than half of the total heritability. In the present study we reconcile these two approaches by showing that linkage signals of height and body mass index (BMI) from 119,000 sibling pairs colocalize with GWAS-identified loci. Concordant with polygenicity, we observed the following: a genome-wide inflation of linkage test statistics; that GWAS results predict linkage signals; and that adjusting phenotypes for polygenic scores reduces linkage signals. Finally, we developed a method using recombination rate-stratif

Lifestyle context

Concrete actions anchored to the cited research. We do not prescribe, we describe.

Diet

  • Balanced diet with portion awareness Moderate

Genetic predisposition to elevated BMI; dietary composition and quantity directly regulate weight

Focus on whole foods and protein; monitor portion sizes and total caloric intake

Exercise

  • Regular aerobic exercise Moderate

Genetic predisposition to ADHD and elevated BMI; aerobic activity improves attention and metabolic health

150-300 minutes moderate-intensity aerobic activity weekly

Lifestyle

  • Consistent sleep schedule and good sleep hygiene Moderate

Sleep deficiency exacerbates ADHD symptoms; variant increases ADHD predisposition

7-9 hours nightly on consistent schedule; avoid screens one hour before bed

Screening

  • Evaluation for ADHD symptoms Moderate

Variant associated with increased ADHD risk; early recognition enables intervention

  • Regular weight and metabolic assessments Moderate

Variant predisposes to higher BMI; monitoring enables timely intervention

Annual weight assessment, metabolic panel every one to two years
