Current genome-wide association studies do not yet capture sufficient diversity in populations and scope of phenotypes. To expand an atlas of genetic associations in non-European populations, we conducted 220 deep-phenotype genome-wide association studies (diseases, biomarkers and medication usage) in BioBank Japan (n = 179,000), by incorporating past medical history and text-mining of electronic medical records. Meta-analyses with the UK Biobank and FinnGen (n = 628,000) identified ~5,000 new loci, which improved the resolution of the genomic map of human traits. This atlas elucidated the landscape of pleiotropy as represented by the major histocompatibility complex locus, where we conducted HLA fine-mapping. Finally, we performed statistical decomposition of matrices of phenome-wid

Lifestyle context
Concrete actions anchored to the cited research. We do not prescribe, we describe.
Diet
-
whole grains and high-fiber foods Moderate
Fiber and complex carbohydrates reduce glucose spikes in those with genetic predisposition to elevated HbA1c
Target 30+ grams fiber daily, emphasize whole grains over refined
Discuss with your doctor
-
personalized diabetes prevention strategy Moderate
rs10047488 C allele shows significant association with elevated hemoglobin A1c; discussion helps establish individualized risk reduction
Exercise
-
regular aerobic and resistance exercise Moderate
Exercise improves insulin sensitivity and glucose control, particularly important with genetic risk for elevated HbA1c
150 minutes moderate aerobic activity weekly plus 2x weekly resistance training
Screening
-
hemoglobin A1c and fasting glucose Moderate
rs10047488 C allele increases hemoglobin A1c levels in large population studies, indicating elevated diabetes risk
Annual HbA1c and fasting glucose testing
